Activity Definition: One unit of Ribonuclease A activity will cause a decrease of 100% per minute in the value of E0 – Ef at pH 5.0 at 25°C.

Appearance: White powder. An enzyme that hydrolyzes ribonucleic acid, sensitive to moisture, and highly soluble in water. Enzyme reaction: Specifically catalyzes the cleavage of the 3′ and 5′ phosphodiester bonds in the ribose portion of ribonucleic acid, forming oligonucleotides with 2′,3′-cyclic monophosphate derivatives. Aqueous solutions are relatively stable below 25°C, with maximum stability in the pH range of 2-4.5. Optimal pH is 7.7. Optimal temperature is 65°C. Isoelectric point (pI) is 7.8. Inhibitors include 0.0005 mol/L magnesium (Mg2?), deoxyribonucleic acid (competitive inhibitor), and denatured deoxyribonucleic acid (more effective than native).
